ARA-290

Research grade peptide

ARA-290 is a synthetic peptide of 11 amino acids, designed as a non-hematopoietic derivative of erythropoietin (EPO). It was engineered to retain EPO's tissue-protective and anti-inflammatory properties while eliminating the blood-cell-stimulating (erythropoietic) effects of the full EPO molecule — meaning it doesn't increase red blood cell count or carry the cardiovascular risks associated with EPO.

Mechanism of Action
ARA-290 works by selectively binding to the innate repair receptor (IRR), a complex formed by the EPO receptor and the beta common receptor (CD131). This receptor is distinct from the classical EPO receptor responsible for erythropoiesis. Activation of the IRR is thought to:

  • Reduce local inflammation

  • Support nerve fiber repair and regeneration

  • Modulate immune cell activity

  • Promote tissue healing and vascular stability

ARA-290 has been studied in several areas, most notably:

  • Neuropathic pain — particularly in diabetic peripheral neuropathy, where small nerve fiber damage contributes to chronic pain

  • Sarcoidosis-associated small fiber neuropathy — clinical trials have examined its effect on pain and quality of life in sarcoidosis patients

  • Metabolic and inflammatory conditions — general anti-inflammatory research contexts

  • Wound healing and tissue repair — due to its regenerative signaling properties

Development History
ARA-290 was developed by Araim Pharmaceuticals, building on earlier work related to EPO's tissue-protective effects separate from its hematologic function. It has been studied in Phase 2 clinical trials for neuropathic pain conditions, though it has not received full regulatory approval for widespread clinical use as of the last publicly available data.

It's a short linear peptide, notable for being one of the first "designer" peptides derived from a larger endogenous protein (EPO) specifically to isolate one biological pathway (tissue repair) from another (erythropoiesis).
